Keyfiles are primarily useful if you want to store the database file somewhere in the cloud you can keep the keyfile offline and manually copy it to the devices you use. What things like the keyfile and Yubikey do is rather splitting up the secret key material that is needed to decrypt the database. Regarding the 2FA question, it doesn't really make sense in the context of a local password database since there is no one to authenticate you. Given that the password DB will reside on my personal laptop and any keyfile or Yubikey I use would be physically in the same place, does 2FA even make sense?
